Big picture...
I'm aware the comic is quite big.
Anyway, the 'breaking' of the fourth wall in a comic is always a tradition for me, so this one is making a joke of myself :P

Adam: The fourth wall is a concept in theatre that a 'wall' exists between the audience and the actors, separating the real from the fictional.
Actions such as addressing the audience are therefore referred to as "Breaking the forth wall."
Therefore, by addressing the audience with "Hello Readers", Mario and Luigi 'broke' the forth wall.
